Point-Mutations in embB306 Gene and Their Association with Resistance to Ethambutol in Mycobacterium tuberculosis in Clinical Isolates
Authors
Abstract:
Background & Objective: Mutations in embB306 gene and their association with resistance to ethambutol (EMB) in Mycobacterium tuberculosis (M. tuberculosis) have not been fully investigated. The aim of this study was to investigate the point-mutations in emb306B gene and their association with resistance to EMB in M. tuberculosis. Materials & Methods: This case (M. tuberculosis resistant to EMB) -control (M. tuberculosis sensitive to EMB) study was performed in the West of Iran (2014-2015), in order to determine the sensitivity of M. tuberculosis strains. Polymerase chain reaction (PCR)-DNA sequencing was used for determining the point-mutations of embB306 gene in both groups (sensitive and resistant to EMB). Data was analyzed by SPSS 16 and Fisher's exact test. Results: Fifty M. tuberculosis strains were isolated from 1019 patients that were suspected to have tuberculosis (TB). 86% of the isolates were sensitive and 14% were resistant to EMB. EmbB306 gene sequencing showed no mutation in control samples; but mutation was observed in 85.71% of resistant samples in case samples. The embB306 mutation showed a significant relationship with EMB resistance (P= 0.00). Conclusion: Mutations in embB306 were observed in the strains resistant to EMB; however, there was no mutation in the sensitive group. There is a direct relationship between these mutations and this type of resistance, so it is an indicator of creation of resistance to EMB in M. tuberculosis.

Mutations in the embB Gene and Their Association with Ethambutol Resistance in Multidrug-Resistant Mycobacterium tuberculosis Clinical Isolates from Poland
Ethambutol (EMB) continues to be used as part of a standard drug regimen for the treatment of tuberculosis (TB). Mutations in the embB gene and those within its conserved EMB resistance determining region (ERDR) in particular have repeatedly been associated with resistance to EMB in Mycobacterium tuberculosis. The aim of this study was to examine the mutational "hot spots" in the embB gene, inc...
full textDetection of embB306 mutations in ethambutol-susceptible clinical isolates of Mycobacterium tuberculosis from Northwestern Russia: implications for genotypic resistance testing.
A total of 183 epidemiologically unlinked Mycobacterium tuberculosis isolates collected in the St. Petersburg area of Russia from 1996 to 2001 were screened for alterations in codon 306 of the embB gene; mutations in this codon are reported to confer resistance to ethambutol (EMB). The embB306 mutations were detected in 14 (48.3%) of 29 EMB-resistant strains and, quite surprisingly, in 48 (31.2...
full textSimultaneous ethambutol & isoniazid resistance in clinical isolates of Mycobacterium tuberculosis.
BACKGROUND & OBJECTIVE There is a need to understand the nature of drug resistance patterns and predictors of emergence of drug resistance in Mycobacterium tuberculosis. There could be common factors/mechanisms for resistance to the drugs, isoniazid and ethambutol, both acting on cell wall. The present study was conducted to analyze the antimycobacterial susceptibility patterns of M. tuberculos...

15 صفحه اولIsoniazid MIC and KatG Gene Mutations among Mycobacterium tuberculosis Isolates in Northwest of Iran
Objective(s) Isoniazid (INH) is one of the main first line drugs used in treatment of tuberculosis and development of resistance against this compound can result in serious problems in treatment procedures. Resistance to INH is mediated mainly by mutation in KatG gene that is coded for the catalase enzyme. The proportional method for detection of INH-resistance is time consuming due to the slo...

Genetic analysis of the embB gene revealed mutations in 17 (68%) of 25 ethambutol (EMB) resistant isolates (M306I, M306V, M306L, Q497R) but also in 4 (20%) of 20 EMB-susceptible isolates of Mycobacterium tuberculosis, namely, an ATG-->ATM substitution resulting in M306I, G406N, and the novel alterations M423I and A659T.
